ABSTRACT

A distribuição espacial e a abundância de carrapatos em estágio de vida livre foram examinadas em fragmentos de Mata Atlântica no Nordeste do Brasil. O estudo foi realizado em Unidades de Conservação de Proteção Integral Mata do Tapacurá e Mata do Camucim, localizadas no município de São Lourenço da Mata, Pernambuco. Os carrapatos foram capturados pelo método da bandeira de flanela em seis parcelas de 200m2, sendo três parcelas estabelecidas em uma borda de mata adjacente a uma matriz agropastoril e outras três parcelas estabelecidas na borda de mata adjacente ao lago de barragem. Foram capturados 2652 carrapatos do gênero Amblyomma, sendo duas ninfas identificadas como Amblyomma dissimile com distribuição espacialmente agregada, demonstrados pela relação variância/média e o índice de agregação de Lloyd. Houve variação significativa na abundância de carrapatos entre as parcelas (H=26,79, p<0,005) sendo encontrados mais carrapatos na borda da mata adjacente à matriz agropastoril. Quanto as variáveis ambientais, houve correlação positiva entre a cobertura de dossel e a densidade de carrapatos (rs=0,31, p<0,05). No presente estudo observou-se que em estágios de vida livre, notadamente larvas e ninfas, a distribuição espacial e a abundância de carrapatos são fortemente influenciadas por áreas mais fechadas das matas. A maior presença de carrapatos próximos à matriz agropastoril poderia facilitar a migração de carrapatos entre ambientes silvestres e ambientes antrópicos.(AU)


The spatial distribution and abundance of free-living ticks were examined in Atlantic Forest fragments in northeastern Brazil. The study was conducted on Integral Protection Conservation Units of Mata do Tapacurá and Mata do Camucim, located in the municipality of São Lourenço da Mata, Pernambuco. Ticks were captured by flagging method in six plots of 200m2, three plots set in a forest edge adjacent to an agropastoral matrix and three plots set in the forest edge adjacent to a dam lake. Were captured 2652 ticks of the genus Amblyomma, two nymphs identified as Amblyomma dissimile with spatially aggregated distribution, shown by the variance/mean ratio and Lloyd aggregate index. There was significant variation in the abundance of ticks between plots (H=26.79, p<0.005) and more ticks was found in the woods adjacent to the edge agropastoral matrix. There was a positive correlation between canopy cover and tick density (rs=0.31, p<0.05). In the present study, it was observed that in free-living stages, especially larvae and nymphs, the spatial distribution and abundance of ticks are strongly influenced by more enclosed areas of forests. Most ticks near agropastoral matrix could facilitate the migration of ticks between wild environments and man-made or modified environments.(AU)

ABSTRACT

Successful programs for ex situ and in situ conservation and management of raptors require detailed knowledge about their pathogens. The purpose of this study was to identify the internal parasites of some captive raptors in Mexico, as well as to verify their impact in the health status of infected birds. Birds of prey were confiscated and kept in captivity at the Centro de Investigación y Conservación de Vida Silvestre (CIVS) in Los Reyes La Paz, Mexico State. For this, fecal and blood samples from 74 birds of prey (66 Falconiformes and eight Strigiformes) of 15 species, juveniles and adults from both sexes (39 males and 35 female), were examined for the presence of gastrointestinal and blood parasites. Besides, the oropharyngeal cavity was macroscopically examined for the presence of lesions compatible with trichomoniasis. Among our results we found that lesions compatible with Trichomonas gallinae infection were detected only in two Red-tailed hawks (Buteo jamaicensis) (2.7%); nevertheless, infected birds were in good physical condition. Overall, gastrointestinal parasites were found in 10 (13.5%) raptors: nine falconiforms (13.6%) and one strigiform (12.5%), which mainly presented a single type of gastrointestinal parasite (90%). Eimeria spp. was detected in Harris’s hawk (Parabuteo unicinctus), Swainson’s hawk (Buteo swainsoni), Red-tailed hawk (B. jamaicensis) and Great horned owl (Bubo virginianus); whereas trematodes eggs were found in Peregrine falcon (Falco peregrinus) and Swainson’s hawk (B. swainsoni). Furthermore, eggs of Capillaria spp. were found in one Swainson’s hawk (B. swainsoni), which was also infected by trematodes. Hemoprotozoarian were detected in five (6.7%) falconiforms: Haemoproteus spp. in American kestrel (F. sparverius) and Leucocytozoon spp. in Red-tailed hawk (B. jamaicencis). Despite this, no clinical signs referable to gastrointestinal or blood parasite infection were observed in any birds. All parasites identified were recorded for the first time in raptors from Mexico. Furthermore, this represents the first report for T. gallinae, trematodes, Haemoproteus spp. and Leucocytozoon spp. in raptors from Latin America. Diagnosis and control of parasitic infections should be a part of the routine in health care evaluations for ex situ raptor populations. El éxito de los programas de conservación de rapaces (ex situ and in situ) requiere de un conocimiento detallado de sus patógenos. El objetivo de este estudio fue identificar los parásitos internos de rapaces en cautiverio en México, así como verificar su repercusión en el estado de salud de las aves. Las mismas fueron estudiadas, decomisadas y mantenidas en el Centro de Investigación y Conservación de Vida Silvestre (CIVS) en Los Reyes La Paz, estado de México. Las muestras de heces y sangre de 74 rapaces (66 Falconiformes y ocho Strigiformes) de 15 especies, juveniles y adultos de ambos sexos (39 machos y 35 hembras), fueron examinadas para analizar la presencia de parásitos gastrointestinales y sanguíneos. Asimismo, la cavidad orofaríngea fue evaluada macroscópicamente para verificar la presencia de lesiones compatibles con trichomoniasis. Lesiones compatibles con la infección por Trichomonas gallinae fueron detectadas en dos (2.7%) Buteo jamaicensis; no obstante, las aves infectadas estaban en buena condición física. En general, los parásitos gastrointestinales fueron encontrados en 10 (13.5%) rapaces: nueve (13.6%) Falconiformes y uno (12.5%) Strigiformes; los cuales presentaron un único tipo de parásito gastrointestinal (90%). Eimeria spp. fue detectada en Parabuteo unicinctus, B. swainsoni, B. jamaicensis y Bubo virginianus; mientras que los tremátodos lo fueron en Falco peregrinus y B. swainsoni. Capillaria spp. fue diagnosticada únicamente en B. swainsoni. Los hemoparásitos fueron encontrados en 6.7% de los Falconiformes: Haemoproteus spp. en F. sparverius y Leucocytozoon spp. en B. jamaicencis. No fueron observados signos clínicos relacionados con la infección por los parásitos internos. Todos los parásitos identificados son reportados por primera vez en rapaces de México. De tal manera que, T. gallinae, y los trematódos: Haemoproteus spp. y Leucocytozoon spp. son reportados en rapaces en Latinoamérica por primera vez. El diagnóstico y control de las infecciones parasitarias deberían ser parte de la rutina de los cuidados de salud de poblaciones de rapaces ex situ. Además, la información obtenida es de gran valor para la conservación in situ de estas aves.

ABSTRACT

Anaplasmose bovina é uma doença com grande importância nas regiões tropicais e subtropicais do mundo por determinar perdas econômicas devido à mortalidade e redução da produtividade. É causada por Anaplasma marginale, uma riquétsia intraeritrocítica obrigatória cujo controle requer, além de uma vacina eficiente, uma acurada identificação de bovinos cronicamente infectados. Apesar de existirem atualmente diversos métodos de diagnóstico dessa riquétsia, os métodos sorológicos, em particular o ensaio de imunoadsorção enzimática-ELISAs, são os mais utilizados devido à sua versatilidade e praticidade. No entanto, devido ao grande número de antígenos disponíveis, atualmente torna-se necessária uma avaliação para definir quais antígenos apresentam um melhor desempenho no diagnóstico da anaplasmose. Soros de bovinos positivos e negativos para A. marginale por PCR, e soros de animais provenientes do Brasil e Costa Rica, foram testados em ELISAs baseados em MSP1a, MSP2 e MSP5 recombinantes, um pool das três proteínas recombinantes, e antígeno de lisado de corpúsculos iniciais da riquétsia (CI). Utilizando soro de bovinos positivos para A. marginale por PCR, uma maior sensibilidade foi observada no ELISA CI. No entanto, uma maior especificidade, com soro de bovinos negativos a PCR, foi observada com os ELISAs recombinantes. O porcentual de bovinos positivos do Brasil e Costa Rica foi maior com ELISA CI. Razões para essas diferenças são discutidas.


Bovine anaplasmosis is a major disease in tropical and subtropical regions of the world by determine economical loss due mortality and productive reduction. The disease is caused by Anaplasma marginale, an intraerythrocytic rickettsia whose control requires, besides an efficient vaccine, the accurate identification of chronically infected cattle. Although the existence of diverse methods of diagnosis of this rickettsia, the serological methods, in particular the enzyme immunosorbent assays (ELISAs), are the most used due to its versatility and practice. However, due to the high number of antigens currently available, an evaluation becomes necessary to define which antigens present the better performance in the diagnosis of anaplasmosis. Sera from cattle positive or negative to A. marginale by PCR, and sera from cattle proceeding from Brazil and Costa Rica, were tested by ELISAs based in recombinant MSP1a, MSP2, and MSP5, a pool of the three recombinant proteins, and initial body lisate antigen (CI). Using sera from A. marginale positive cattle by PCR, the highest sensitivity was shown by CI ELISA. Nevertheless, the highest specificity, with sera from negative cattle by PCR, was shown by recombinants ELISAs. The percentiles of positive cattle from Brazil and Costa Rica were higher with CI ELISA. Reasons for such differences were discussed.

ABSTRACT

Antigenic characterization of Anaplasma marginale isolates, by identifying conserved and variable epitopes of major surface proteins (MSP), is an important tool for vaccine development against this rickettsia. The B cell epitopes of A. marginale isolates from three microregions of the State of Pernambuco and one from the State of Mato Grosso do Sul, Brazil, were characterized by indirect fluorescent antibody technique (IFAT) and Western blot (WB) with 15 monoclonal antibodies (MAbs). The epitope recognized by MAb ANA22B1 (MSP-1a) was conserved by IFAT and WB (73-81 kDa). MSP-2 epitopes recognized by MAbs ANAO58A2 and ANAO70A2 were conserved by IFAT, while ANAO50A2 and ANA66A2 epitopes were polymorphic; in the WB, the MAbs ANAO50A2 and ANAO70A2 identified bands of 45 kDa only in the Pernambuco-Mata isolate. None of the isolates reacted with MAb ANAR75C2 (MSP-3). The MSP-4 epitope recognized by MAb ANAR76A1 was conserved by IFAT, as well as the MSP-5 epitope recognized by MAb ANAF16C1 by IFAT and WB (16 kDa). The MAbs ANAR17A6, ANAR83B3, ANAR94C1, ANAO24D5 and ANAR19A6 identified conserved epitopes by IFAT. MSP-1, MSP-2 and MSP-4, which previously showed partial protection in experimental trials, are also potential immunogens to be employed in Brazil, due to the B cell epitope conservation

Este estudio tiene como objetivo, caracterizar el estado coproparasitológico de los canarios (Serinus canarius) mantenidos en cautiverio en el estado de Pernambuco, Brasil. De 327 canarios examinados, 167 (50,5 por ciento) presentaron infección por Isospora.
